because of the original method .. there is a chemical reagent that is very difficult to get and use in my country .. or I need special permits to use it .. greetings!
Uranyl Acetate is used to determine the rotation of a corn starch/corn sugar solutions. I doubt, very much, your concentration is high enough to determine the amidase activity. You can try reviewing ICUMSA (International Commission of Uniform Methods of Sugar Analysis) methods.
You might try incubating a known concentration of starch in the soil and perform a back titration with Standard Iodine Solution (use KSCN to sharpen the endpoint).
