May I ask you some question? I highly need your help
I want to infect human foreskin fibroblast with CMV standard strain(AD169) to study NK cell activation with that ,unfortunately, we can 't obtain it(because of biosafety 2 and Prohibit to import Iran ) I highly need your help:
1-How can I infect this cell line? Can I use CMV lysate for this aim instead of the live virus? Or
2-if I use PBMC from patient with reactive CMV infection as an infected cell line for stimulating isolated NK cell from a healthy case or PBMC of the patient with active CMV (high copy number of CMV genome ) can infect fibroblast cell line?

I am working on CMV, You can isolate your own CMV strain from CMV positive patient's saliva, then passage it many times in fibroplasts to get attenuated virus strain so you can work with it
