I ordered the standard oligo with no modifications. I am aware in order to tag with Cy3, there needs to be an amine group. Is there in such a way I can modify the oligo so that the Cy3 tagging is possible?
Yes. A lot of fluorescent dye labeling relies on simple reactions such as Click chemistry or maleimide chemistry for protein labeling. For DNA, our lab uses an nhs ester amine reaction to site specifically label a DNA substrate containing a nucleotide base modified with an additional amine functional group. You can order DNA with these modifications directly from IDT and get free dye from various sources.
