I have a steady state reaction of an enzyme involving 2 substrates. I have the velocities and the 2 substrate concentrations. I need to know the type of the reaction that is catalyzed, i.e. whether it is a ping pong or ordered sequential. How can I fit the values of velocity for each set with the changing concentrations of the substrates to do a global fit to get the values of Vmax, Ka and Kb for the overall reaction?
That will be good, but i need to use a program like origin or graph pad to do it

You can understand the nature of the reaction, i.e. whether it is a ping pong or ordered sequential, by knowing the reaction mechanism. I think it is very simple if you are familiar with the mechanism and if you draw the CLELAND diagram, it will be very easy to understand. Once you know about the mechanism, just continue with the kinetics as suggested by Uttam Pal. You can draw the velocity curves in Origin or graph pad or even in excel and the curve fitting models can be obtained.
