I have a question about a wnt signaling pathway. I want to know how exactly a Wnt–FZD–LRP5/6 trimeric complex is formed. Whether WNT3a (activator of LRP6) and Dkk1 (inhibitor of LRP5/6) have a competition to interact with LRP6? And if Wnt can bind with LRP6, the active site of DKK1 (that is overlapping or very close to Wnt active site on LRP6) is occupied and DKK1 can no longer bind with LRP6 or is it possible even after the formation of trimeric complex (Wnt–FZD–LRP5/6) DKK1 can bind with this complex and inhibit the LRP6 and finally the pathway is inhibited.

The other question is whether any data about the interaction of the first two B-propeller (B-propeller 1, 2) of LRP6 with the other proteins specifically Wnts are available or not?
