When I want to transform a plasmid into three Xanthomonas translucens pv. undulosa, found there is one WT resistance to Ampicillin, I tried purify plasmid and transform into E.coil, but I got nothing, so I think this strain(WT) naturally resistance to Ampicillin, is there any expert know about the meaning, please give me some advice?
There are often natural isolate which are either naturally resistant or tolerant to antibiotics. You can easily test if this strain is producing beta-lactamase and degrading added ampicillin. If this is the case, then you probably can't use that strain. However if the strain is just tolerant to Amp, then you can try using the drug at a much higher concentration. When I was working with Serratia marcescens, it was pretty tolerant to antibiotics (but not actually resistant). By using ampicillin at 500ug/ml instead of 50, we were able to select for transformants.
