I am also looking for some good cell penetrating peptides to deliver oligos systemically; both of which are to be delivered by I.V. (If possible please include sequences.) I am aware that specific tumor types may have different optimal cell penetrating peptide sequences, however taking advantage common phenomenon like glutamine addiction may help in their common design, as well as known tumor targeting peptide sequences. Cell penetrating peptides that target to specific organ sites may also prove useful. Please include any relevant toxicity information if available.
I want to use them for morpholinos and want a CPP that can preferentially concentrate them to tumors to reduce waste and lower cost I have some design ideas similar to TAT but was wondering if there was anything better for targeting to cancer cells specifically that others have used successfully.
What is not clear is if you want to form complexes or just a chemical conjugation between CPP and ON. Going for a morpholino, I guess you have chosen the conjugation route...
Having said that, there is no "classic" receptor for CPPs. We wrote a review last year about the role of GAGs in the uptake of CPPs (and virus and poly/lipoplexes). Have a look at it, you may find it useful. There are also several papers that describe the ideal design of a CPP, but I think you are already aware of those.
