I would use peripheral blood and spleen together to obtain a good amount of monocytes. These cells can be isolated according to their Ly6C expression after depleting Ly6G neutrophils which are also Ly6C positive. CD115 is an alternative marker to select mouse monocytes. There are MACS kits for this purpose.
