I am confused that whether that protein will be secreted into media and I have to use that media for further experimentation or will it be remain attached to cell surface and I have to centrifuge cells to get my protein into supernatant?

In our case, most of the proteins were secreted into culture medium. You can directly collect the medium and cfg 2,000g 5min (to get rid of the cell debris). The sup can be used for further experiments.
Just be careful when using this for western blot cause it contains lots of FBS, which may severely affect your results.
