You need to use freshly prepared, clear yellow streptozotocin within a few minutes of preparation because it has a rapid oxidation and gets cloudy within about 10 minutes. Make up only the amount you need and use in 10 minutes.
if you are doing many rats make more than one aliquot and do not add the solvent until you are ready to inject. I t is most important to give intravenous Streptozotocin to get uniform levels of beta cell destruction and diabetes.
I have rats with indwelling catheters and can give intravenous injections very quickly and get excellent diabetes every time. = glycosuria in first 24 hours.
what is the dose of streptozotocin you are using and what route are you using to inject? Intraperitoneal injections can work but unless you know how to avoid gut shots where the injection goes into intestinal lumen you will have variable and disaapointing results. I have never used alloxan and streptozotocin in combination so cannot answer your question with any clarity. Do you give streptozotocin to fed or fasted rats?
perhaps try the combination and see what happens. The route of administration is also very important and you did not answer when I asked about the route???
I have only used intravenous route because I wanted a uniform severity of diabetes.
Intraperitoneal injections are only effective if you can avoid doing a gut shot where the injected material goes into intestinal lumen instead of into the peritoneal space. using a l very small guage injection needle will most likely give poor results because the injection goes into intestines most of the time.

Nevertheless, as I mentioned, this might be the first time anyone has heard of such an attempt.
Suppose you use alloxan and streptozotocin to induce diabetic mice in the subsequent biochemical tests (IL-1, IL-10, etc.). In that case, you should compare the experimental results of alloxan or streptozotocin alone or the model animals used previously. The regulated model animals are used for comparison in the hope that the data you receive will be of reference value and reproducibility.
