I want to run hplc analysis using tannic acid as standard. May I know whether ACS reagent is applicable for hplc analysis. I kept searching for tannic acid hplc grade but could not find one. Anyone here has experienced doing hplc using tannic acid. Mind sharing about the difference of ACS and HPLC grade. And where can I specifically get the hplc grade tannic acid. I have searched through Sigma and Fischer but they did not mention whether the product is applicable for hplc or not.

Quick answer: ACS Reagent grade chemical are often a bit "dirty". "HPLC Grade" refers to the quality of the mobile phase ingredients and solution, not the sample. ACS grade is a "clean" but crude grade for most reagent chemicals. It assures you of receiving the right chemical in a purity good enough for routine bench work. No one would probably sell or offer Tannic acid in HPLC grade as it is not commonly used in HPLC work. As a sample (or std), you would be using it chromatographically, in a very small amt as a retention time and spectral marker, but do not expect one peak (it will be a mixture). You will be fine using it as a marker, but not as a "standard". Be sure and filter the Tannic acid final stock std solution through a 0.45 or 0.2 u filter before placing in a vial for use with the HPLC system.

"ACS reagent" grade tannic acid is actually a complex mixture of polygalloyl glucose species extracted from plants. (The "reagent grade" designation merely means that it's "good enough" for laboratory use.) HPLC with good resolution will show it to be a mixture, so it's far from ideal as a standard. You'll have to isolate and purify a single component if you require a clean peak in your chromatograms. The attached pdf describes a procedure for isolatiing pentagalloyl glucose.

Analyzing tannic acid using HPLC?..You have to first of all check for the mobile phase that will be suitable for running tannic acid and a suitable diluent which could be a mixture of solvent. after dissolving the ACS grade tannic acid in the diluent, if it is completely soluble fine but if not you can also sonicate the sample using an ultrasonicator to aid its solubility and filter using 0.45um whatman filter paper...you can refer to British pharmacopeia (BP)vol.1 or 2.
The difference between ACS and HPLC grade solvent is that the ACS is for normal laboratory analysis with some level of purity but the HPLC grade is simply very pure not crude and free from any Dirt.. pure enough not to have anything that could block the HPLC column and the chromatogram will be fine with clear and single peak... but from my experience in operating HPLC wen we run out of HPLC grade solvent.. I always use ACS grade but will always filter using a vacuum pump then degas using a sonicator to remove anything that could block the pumps or column... so to answer ur question... Yes!! ACS grade can be used for HPLC but filter and degas the solvent to avoid column and pump blockage and also to get a fine peak with clear chromatogram.. I hope this help!! all the best!!....
