Dear Friends, Please suggest me , how i can calculate resolving power and allele frequency of a molecular marker (RAPD/ISSR). Is there any software to calculate these parameters?
Resolving power (Rp) which shows the ability of the most informative primers to differentiate between the genotypes was assessed according to Prevost and Wilkinson, (1999) using:
Rp = ∑ Ib
where Ib is the band informativeness with Ib= 1 - [2 x (0.5-p)] and where p is the proportion of clones containing the band. The resolving power is based on the distribution of detected bands within the sampled genotypes.
Discriminatory power of each marker (RAPD and ISSR) it could be evaluated by means of three parameters, viz. Polymorphic information content (PIC), the probability in detecting polymorphism by a primer or primer combination between two randomly drawn genotypes was calculated using the formula PIC = 1-Σpi2, where pi is the frequency of the ith allele (Sehgal et al. 2009); Resolving power (Rp) which is the ability of each primer to detect level of variation between individuals was calculated according to Prevost and Wilkinson (1999): Rp = ΣIb where Ib (band informativeness) takes the values of: 1–[2|0.5–p|], where p is the proportion of individuals containing the band, and marker index (MI) for each primer was also calculated as a product of two functions - the polymorphic information content and effective multiplex ratio (EMR) (Varshney et al. 2007) i.e., MI = PIC × EMR.
"p" in lb= 1 - [2 x (0.5-p)] formula is the proportion of the genotypes containing the band. I hope you will come to your question by reading the following article.
