Dear colleagues

I plan to run an experiment aimed to test the biocompatibility of some material. To do so, I plan to check adhesion and proliferation of murine osteoblast precursor cell line MC3T3 to the tested substrate.

My question is: how to culture the cells for this experiment? According to ATCC, MC3T3 must be cultured in aMEM without vitamin C in order to keep them undifferentiated. Most of papers test MC3T3 in the complete aMEM (thus, the cells probably moved towards osteoblastic differentiation).

I am a newbie in the field, so I wonder, whether it is critical to test cells on the material when they are undifferentiated or when they moved towards osteoblasts.
