Hello everyone,
I am currently trying to stably transfect the pancreatic cancer cell lines Panc-1, BxPC-3, AsPC-1 and MIAPaCa-2 with plasmid DNA having a G418 resistence gene.
I tried FuGene HD with different reagent to DNA ratios (2:1, 3:1, 4:1) always using 1 µg DNA and 4 to 8 µl of FuGene HD in a well of a 6-well plate. Real-time PCR shows that my gene is expressed but when I trypsinate the cellls and seed them into dishes to select them with the antibiotic, they all die after about 10 days (I exchange the medium and G418 every 3 days).
Now I try to use ViaFect as well, but nobody in my lab has used this reagent before.
Maybe some of you have worked with these cell lines before and can help me? I saw some answers using cell specific reagents from altogen, but I would like to try it with a more common reagent because I am the only one working with those cell lines.
