How to test for Ca(2+) free versus Calcium bind to Calsequstrin?

Hi,

I was wondering if anyone would know of a simple method to measure the calcium free in Sarcoplasmic Reticulum and/or the calcium bind to Calsequestrin in mouse adult cardiomyocytes.
