I have isolated two gram negative bacterial strain and would like to estimate biofilm formation capacity. However, these strains are known for biofilm formation, therefore i would like to take one of positive control that can help us to determine the biofilm forming capacity.
I suggest to use a standard bacterial strain as a positive control (e.g. from the ATCC collection) and a culture medium without bacteria as a negative control.

S. aureus ATCC 25923 and S. aureus ATCC 35556 are used as negative and positive controls, respectively, for the biofilm assay.
thank you Krzyzek & Elmanama. I would like to ask a suggestion actually i don't have these cultures as you have suggested but i have ATCC 25922 (E.coli) and ATCC 43300 (S.aureus )culture will it be fine to use ATCC 25922 as a negative control and other one for positive control.
Both strains in this case will be positive controls. Only the medium without the presence of any bacteria can be taken as a negative control.
