We normally used to store them in the solution of glycerol with plastic beads. So inoculate SAB plate and then use a sterile cotton swab to gather some colonies. Dissolve colonies in glycerol solution and shake the eppendorf tube. Get rid of liquid using plastic sterile pipette. So you have beads covered with Cryptococcus to store. Store it in -80. All yeasts can be stored this way for many years without any problem. Later on you thaw the eppendorf and drop few beads on the agar plate. Leave the plate to incubate for 1-2 days in 37 degrees. From this you can use colonies to streak another plate to get some more colonies. Really simple and straight forward. Good luck.
