I am trying to find a way to visualize the plasma membrane in tissue sections prepared of different mouse organs in confocal microscopy. The aim to separate the intracellular and extracellular compartments in image analysis. We tested fluorescent phalloidin that nicely stained the subcortical actin filaments in the liver, but was much less usable in other tissues. I'm afraid of lipid-specific stains. They are good for cultured cells, but I guess they would stain everything in tissue sections. I am considering fluorescent lectins, but I haven't yet tested them. Does anyone have any experience in this problem? Which lectin would you recommend?
