I need to perform an IC50 test of L-glutamate on a SH-SY5Y cell culture, but I'm having some issues with diluting this L-glutamate. I'm using L-Glutamic acid (Sigma: G1251), and in the literature, toxicity assays with L-glutamate are tested at concentrations between 20-100 mM, which I consider quite high. My difficulty is that the product's technical information states that the solubility is 100 mg/ml in 1M HCl, but it is not feasible to use HCl in a cell culture. I've tried to solubilize it in water and DMSO, but it is not dissolving. Another question I don't understand is how they manage to obtain a concentration of 100 mM since this compound has a molar mass of 147.13 g/mol, and to make a 1M solution, for example, it already exceeds the compound's maximum solubility.
