The gastric cancer cells have been implanted subcutaneously into nude mice previously. I want to seperate the metastatic cells, but the cells don't have any protein markers. Can I use some cell surface markers such as HLA or CD to identify human cancer cell and mice cell by flow cytometery? If I can, which CD could be more specific and efficient?
HLA A-B-C and ß2microglobulin should work fine. You have to carefully check for cross reactivity.
It seems you would like to get gastric cancer cell in cell suspension. If that is the case, I agree with Dr Ayata`s proposal. However, I should point out that metastatic tumor cells often down regulate Class I antigen for example. Thus, you should be careful about it. If you aim is achieved from tissue sections, make serial frozen sections and stain one section with anti-human vimentin antibody. Then you can micro dissect metastatic human tumor foci from sections.
Do your tumor cells have uniform expression of human epithelial cell markers - ie human CD24? You could add the mouse H2-q as a negative channel perhaps.
