Hi Omar! The column is a Eclipse Plus C18 (3,5μm, 4,6x100mm), Agilent. The sample has a high sugar concentration. We believe that something blocked the column and lead to a backflush of the solvents applied. Then we deteted a leak on the pump. Just before the system shut down due to the leak we noticed a peak on the DAD of about 70000 (several dozens larger than the largest we detected with the standards). We are concerned about the column. It was running on TFA (0,1%) and acethonitrile. Thanks in advance!

Rinse the column with water with no organic modifier like acetonitrile, that should elute the hydrophilic components like sugars.
