According to several published papers，I added 400 mM HFIP in the mobile phase to increase the intensity of oligonucleotides in LC-ESI-TOFMS. However, the intensity of HFIP (m/z 166.9983 [M-H]- and 344.9953) were so strong that I can not obtain any signal of my target. Could you give me some suggestion about how to reduce the interference of HFIP?

Just a word of caution. Does your LC/MS (HPLC) system use an in-line vacuum degasser? If so, the use of HFIP may not be compatible with your degasser and result in serious damage and contamination to the entire system. Some vacuum degasser chambers are compatible and some are not so please verify before you continue to use it (My company repairs a lot of damaged degassers which were used with fluoroalchohols. HFIP is usually not recommended with membranes made from Teflon AF).
