I am using acetonitrile: water (40:60) as a mobile phase for making the dilution for HPLC analysis. The solution has quick degrade by taking HPLC analysis. Please suggest me how to reduce degradation rate of berberine chloride in mobile phase ?
The question is it is degradation in time of the analysis or the "degradation" occurs in the sample vial, which is more likely. I would go for something like an antioxidant in the sample solution, lower the autosampler temperature or change to plastic vials, which prevents for adsorption of substance on the glass wall.
Adam Zmysłowski I have tried to prepare BBR dilutions in mobile phase (ACN:water) or methanol and take BBR peak in HPLC. Every time dilution was degraded gradually and we are unable to plot BBR standard.
Berberine chloride is photosensitive and thermosensitive. So try to prepare the solution in dark, also keep the temperature below 25'C before and during use.
