Hi there,

I guess the DNA is actually a plasmid. If so, elute the DNA using 10-20µL of sterile water or TE and perform transformation (using the eluate as DNA source) to obtain clones from which you'll be able to prepare DNA miniprep.

You can recover DNA from filter paper by cutting out the marked circle area that contains the dried plasmid DNA and immersing it in TE buffer.

To recover the plasmid, use a clean razor blade toof the cining your DNA.

Immerse the circle in 30µL of TE and pipette to mix.

After waiting for at least 10 minutes, use 2µL to transform competent bacteria.

Hello, to recover the DNA you must put the filter paper with a small volume of water or TE and leave it for about 20 minutes, then recover the water and the DNA will be there.
