i need to separate Anaplasma marginale from infected blood. I search many literature and research paper but not getting a sure shot answer. pls help me in this matter.
DNA was extracted from whole blood using the High Pure PCR Template
Preparation Kit (Roche) following the manufacturer’s instructions. Briefly, 200 μl of
blood was mixed with 200 μl binding buffer and 40 μl proteinase K, incubated for 10
minute at 70 ◦C. Added 100 μl of isopropanol and sample loading onto a column. This was followed by successive washing, then collect DNA in a final volume of 100 μl.
Feel free to contact me at any time if you has any questions in the protocol
if what you want is to purify the initial bodies form infected blood I can send you our protocol which includes freeze-thaw, hipotonic lisis, sonication and gradient centrifugation to obtain 99.99 membrane-free initial bodies.
