I am struggling at finding a suitable approach for extracting soybean seed’s storage protein fractions, i.e., albumin, globulin, prolamin, and glutelin, and their subsequent determination through UV-Vis Spectroscopy using Bradford staining solution and bovine serum albumin (BSA) as standard.

As the obtained results were significantly different than those found in the literature, I came across some major issues:

1) Is the sequential extraction (water, sodium chloride 5%, ethanol 60%, and NaOH 0.4%) method suitable for extracting the albumin, globulin, prolamin, and glutelin fractions of soybean seeds singly?

2) Is the BSA an adequate standard for determining either globulin, prolamin, or glutelin fractions?
