I tried forming chitosan films using some methodologies I read from certain journals. I used 1% (v/v) acetic acid to form a 1% (w/v) chitosan solution which was stirred overnight and was filtered through a cheesecloth. After filtration, I sterilized the solution at 121 deg C for 15 mins, cooled and then added 0.50 mL/g glycerol as plasticizer. I poured the solution on open glass petri dishes and dried it at 40 deg C for more or less 4 hours. The films were too adhered on the glass plates that when I tried peeling them off immediately after drying, I only got a little film. Some wouldn't peel off. I tried scratching the surface of the plates just to make sure films were formed and that the chitosan solution didn't evaporate at all. Did I do something wrong? What methods can you suggest to improve my solution preparation and casting method? I am currently trying casting on disposable plastic plates with the same chitosan solution and hoping this time will be successful. Thank you so much!
