I am going to collect human blood from remote areas (10 hours drive from my lab) and extract PBMC in my laboratory. The place where I collect the samples has poor lab facilities. Thus, I need to keep PBMC alive, while transporting the samples to my lab. Does anyone have any suggestions, ideas or any particular methods I could use to maintain the quality of the samples?
I have been working on stability of fatty acids (FA) from human blood derivatives stability (specifically plasma, serum and red blood cells) under different storage conditions (temperature, antioxidant, thawing) and after a year we found not variation in FA at -80 degrees in samples contining antioxidant. In addition, samples at -20 were equally stable for several months. Unfortunately have not worked with whole blood.
Frozen human blood remains stable for several months I've been extracting DNA from human blood frozen for several weeks and everything went really fine... Keep in mind that you're going to need a real good refrigerator capable of freezing your sample at least -30 Celsius or close... good luck!
I suppose you collect the blood on EDTA tubes, then keep them cooled for transport:The samples must not be frozen ! isolate PBMC next day or as soon as possible
You should use citrate instead of EDTA to improve PBMCs viability. 1-2 days of delay until isolation is not a problem. Mantain at RT.
