I am working with gold nanoparticles tagged with Alexa Fluor dyes and need to injected them into mouse tumors. I would like to visualize their internalization and distribution within the tumor tissue using confocal microscopy.

Could anyone please advise on:

  • The best way to preserve tumor tissue post-injection to retain Alexa Fluor signal?
  • Whether paraformaldehyde (PFA) fixation (e.g., 4% PFA perfusion and post-fixation) affects the fluorescence intensity or stability of Alexa Fluor dyes in tissue, especially when conjugated to nanoparticles?
  • If anyone has experience with similar nanoparticle imaging protocols (e.g., fixation time, cryoprotection, or sectioning method), I would greatly appreciate your input.
