I want to do TEM of nano-particles treated fungal cell to localize the nanoparticles within it. Is there any special requirement or precaution that we should take while doing TEM of cells. Please suggest me some operating condition. such accelerating voltage etc.
It is challenging job to prepare a good sample for high-resolution electron microscopy of polycrystalline ceramic powders containing hard particles. a fungal wettable powder is prepared by using 11.0% of carboxymethyl cellulose-Na, 9.0% of polyethylene glycol 6000, 5.0% of primary alcohol ethoxylate, 2.5% of glycine, 5.0% of fucose, 27.5% of kaolin and 40% of freeze dried fungi by response surface methodology (RSM).
You may prepeare as like as for bacteria, fix the fungus, stain and proceed.
Hi Mohd Qasim, I would prepare your fungal cell as you would for normal cell. The process will take around 5 days and a bit of luck. You will need to fix the cell, then heavy stain it with OsO4, dehydrate and then place the samples in resin for microtome. Please private message me and I can send you a copy of my procedure.
