Catechin is not the best for calibration curve as Philip noted above, rutin will give a better result. For the preparation dissolve 5 mg of catechin in 5 ml of analar methanol to obtain a stock concentration of 1mg/mL, then prepare 20, 40, 60, 80 and 100 microgram per mill. Use these concentrations to prepare the standard curve.
The standard catechin solution to be suck into pipette (0.25 ml, 0.5 ml, 0.75 ml, 1 ml, 1.25 ml, 1.5 ml, 1.75 ml & 2 ml) and transfer into respective conical flasks, labelled as 5 ppm, 10 ppm, 15 ppm, 20 ppm, 25 ppm, 30 ppm, 35 ppm, 40 ppm and 45ppm respectively. Then, conical flask filled with methanol until 1ml for each flask individually.
