I recently purchased the MCE compound Artemitin (Artemitin), item number: HY-N3017, which belongs to the flavonoids. I would like to ask how to dilute the product in cell experiments? At present, I have used DMSO to dissolve the compound into a mother solution with a concentration of 50mM. My working solution concentration is 400μM, 200μM, 100μM..., and precipitation occurs during the dilution process. I have tried heating in a 60℃ water bath and ultrasonic for 30min, but there is no way to completely dissolve it. White floccules can be observed with the naked eye, and needle-like crystals can be seen under the microscope.
Three dilution methods have been tried so far:
1. Directly dilute with a mother solution concentration of 50mM to the working solution concentration.
2. Dilute with a 50mM mother solution concentration to 10mM, then dilute with 10mM to 1mM, and finally configure the working solution concentration with 1mM.
3. Dilute the 50mM stock solution to 5mM and prepare the working solution at 5mM.

Since you've already prepared a 50 mM stock solution in DMSO and are observing precipitation at lower working concentrations (400, 200, 100 μM, etc.), the problem likely stems from the way the compound is being diluted into aqueous media. Flavonoids often have poor water solubility, and when the DMSO content drops too quickly during dilution, the compound can precipitate out. One helpful strategy is to perform a stepwise dilution: first dilute the 50 mM stock into 10 mM and then to 1 mM, all in pure DMSO. Only after that should you make your final working concentration in cell culture media—ideally pre-warmed to 37–40 °C—and add the DMSO solution drop by drop while gently mixing. It’s also important to keep the final DMSO concentration consistent (typically 0.1–0.5%) across all samples, including controls, to avoid variability and precipitation. If precipitation still occurs, you could try using solubility enhancers like a very low concentration of Tween-80 or PEG-400, or consider adding serum if your media allows for it. Lastly, if visible precipitates remain and can't be avoided, you might filter the final solution through a 0.22 μm syringe filter, though this may slightly reduce the effective concentration. Always prepare fresh working solutions before each experiment, as flavonoids are often unstable in solution over time.
