What is the method you are using for amylose determination?
You will need to have a source of pure amylose. It is best that the amylose standard be from the same botanical source as the starch you are analyzing. This is especially true if you are using an iodine affinity method since the color produced will be different depending on the source of starch.
You will need to dissolve the amylose in the solvent you are working with for your assay at a concentration which is higher than the maximum amylose content of your starch samples. The you can prepare several dilutions and the lowest concentration should b lower than the minimum amylose content for your samples.
