Are you asking how to obtain a chromatogram from a column without an HPLC and its associated detectors? Are you doing chromatography without an instrument such as an FPLC or HPLC that automatically collects the chromatogram in real-time from its detectors? If so, then you will have to collect many fractions of eluate of equal volumes and measure the amount of protein in each one, either by a protein assay or by UV absorbance. Then use any graphing software to plot protein concentration or UV absorbance versus elution volume. That is your chromatogram.

What do you mean with elution volume?  each fraction's volume or total eluted volume?

Total eluted volume. For example, if you collect fractions of 1 ml each, fraction 26 consists of the 25th through 26 ml, fraction 27 consists of the 26th through 27 ml, etc.

I agree with you, and I do not like computer (PC; personal computer)-aided recording method, which has been integrated into the HPLC system at all.

Observating the real-time chromatogram is surely the peace and content to work with HPLC machine. 

Therefore, please use the pen-type recorder for HPLC.

Chromatopac (Shimadzu) is convenient to subtract the gradient base-line (or gradient blank; see file PTH Roma for Chromatopac C-R6A).   Recently, I have used a new version of Chromatopac in the fucoidan determination ( see file JCB Okinawa for Chromatopac C-R8A).

Signal information is available from the back-side panel of photometric detectors (out-put at 1 V).   Signal line or cord is attached to recorder.

I'm surprised to learn that pen-type HPLC chart recorders are still available commercially. I thought they had been replaced by computers at least 20 years ago.
