Want to reduce the Hardness of TPU. As TPU comes in a granular form incorporating plasticizer is difficult. What methods can be used to incorporate plasticizer in TPU?

Yes, and the best plasticizer is the one of polymeric type such as PLA, to avoid the migration problem. I think one of the suggested document deals with this option. My Regards
Traditionally, waxes have been used to plasticize TPUs which also comes with blooming problem. Either reactive plasticization or selection of softer molecules as the building block of TPU would give you the target softness.

They are not toxic and of low fugability. They are most often used for PVC but should also work for TPUs. They are easier to introduce in the TPU than oligomeric plasticizers. Perhaps even simple swelling before injection molding is sufficient.
