I have designed an SPR based PCF sensor, which has a TiO2 and Gold coating. A reviewer has asked for the characterizations (e.g. SEM, TEM) to study the surface morphology. The reviewer has also asked to establish the effect of morphology on efficiency. How to perform these things? Can anyone help me out?

SEM and TEM are basically surface characterization techniques to determine the morphology and particle sizes of materials. The main difference between the two techniques is that in SEM the images are produced by detecting reflected or knocked-off electrons, while TEM uses transmitted electrons (electrons that are passing through the sample) to produce the image.
Since BOTH samples are conductive, coatings is perhaps not needed. You just need to run the analysis using the instruments and determine their respective morphologies. Also from Energy Dispersive X-ray (EDX) analysis of the SEM will give you surface elemental compositions of the samples.
A beam of electron strikes the material, then SEM takes image by detecting of reflected or knocked-off electrons. while TEM create image from electrons that are passing through the sample. Transmission electron microscopes are capable of imaging at a significantly higher resolution than SEM. smaller de Broglie wavelength of electrons of TEM to capture fine detail, even as small as a single column of atoms, which is thousands of times smaller than a resolvable object seen in a light microscope.

You study the surface morphology, compare the surface roughness, or any other special characteristic features that are seen on your modified catalyst that can be reported. You need to compare Au-TiO2 to only TiO2 and study the surface characteristics and come to a conclusion, how the surface morphology has helped in you sensing? TEM will help you to study the size of AuNPs as well as the diffraction, which will provide some more details of your composite.
