I have some polysaccharide fractions obtained by sepharose CL-6B column separation. i wish to determine its approximate molecular weight by carring out SDS PAGE. if anyone know, how to do this?
Sorry, but SDS PAGE is not the best way for this. Polysaccharides are polydisperse, and will not give a sharp band on PAGE. In particular, the SDS will not bind in the same way that it binds to proteins, especially with a sulfated polysaccharide. I suggest you use HPSEC and/or light scattering for a determination of Mw and distribution.
