I would like to measure the total iron (even the bounded one) in my bacteria.
I want to measure it by EPR.
I think that I have to completely destroy the cells and then add a iron chelator.
In the paper "Use of Blue Agar CAS Assay for Siderophore Detection" 2011, they used hydroxyquinoline and chloroform to remove iron from casaminoacid solution, I could do the same on my bacteria, which would kill them and i could have the iron in chloroform (then, dry the chloroform to have the total iron).
I don't know if this method would be the best one, maybe use deferoxamin on sonicated cells...
If someone has a better idea, that would be very great!
