I need to measure the thickness of a semi-clear membrane (about 100 um thick). Ideally, thickness would be measured without touching it! Help would be greatly appreciated!
(1) SEM : You can clearly see the thickness by the SEM. The sample for SEM analysis of the polymeric membrane was prepared by fracturing it in liquid nitrogen. It should be dry under vacuum before analysis. For non-conductive sample (Polymeric material), you have do sputter coating of the gold layer (thickness 5-15 nm ) on the sample before analysis.
(2) Gravimetric method : It is used to measure the effective thickness of top layer material (on support).First we have to measure weight gain of the sample after coating. Use the following equation to apply this method.
Total thickness = (Wf-Wd)/(Density of coating material * surface area of support )
Where, Wf and Wd are the weight of coated membrane and weigh of uncoated membrane respectively
The gravimetric method also give the idea about effective thickness of top layer material by measuring the percentage of the intruded mass of the top layer into the pore of the support layer .We can also use this method for self-supportive membrane (without any support)
