In fermentation when the broth is gradually becomes viscous what will be the most relevant method for biomass measurement. as it becomes difficult to separate the actual biomass from the other metabolites produces in the broth .
The answer below are good It depend of the nature of your ferment.
Plate count with a specific media for your microorganisms should be good. Any Indirect measurement could be possible (deshydrogenase, ATP, respirometry, CO2 production etc.) with a standard curve can help to estimate biomass activity and or number. Also, if It's possible you can wash (dilution and centrifugation with supernatant removal) your biomass in a saline to remove metabolite or viscous fraction.
It's difficult to propose THE best method. It depends of the nature of the biomass, media etc. It's better to try 2 or 3 methods and choose the most accurate for your case.
