I followed several papers using ammonium molypdate method but there was precipitation and the results were inconsistent. The protocol is as followed:

- Extract freeze-dried leaves with HClO4 10%, then neutralize excess acid with KOH/triethanolamine and centrifuge.

- Add 200 ul supernatant to 20 ul of reagent including ammonium molypdate, ascorbic acid, sulfuric acid and potassium antimonyl tartrate. Wait 15 min and read OD at 885 nm.

The reagent reacts fine with Na3PO4 solution but there was precipitation when added to the sample. How do I solve this?
