hello! I'm synthesizing Gold Nanorods and I get both peaks with UV-Vis and all the colour changes go as per mentioned in the literature with seed-mediated synthesis. But, I never get the results in SEM analysis. We do not have the facility of TEM, so I need help for preparation in sample for SEM. Kindly guide me from the point when I have the sample in suspension with the violet color developed. How should i transfer to glass slide to dry it ? Guide me about centrifugation and dilutions and if available, any other protocols. I always get the sheets or layers in SEM analysis and some nanoparticles occasionally and not any rods.

Hi Mudassir Khan , you need to dilute the sample before preparing for SEM. I am thinking, may be the sheets or layers you are seeing in the SEM could be the rod but in high concentration. May be that is why you are unable to see the morphology. What I propose is, try to dilute your solution, let say, take an aliquot ( eg. 20 micro litre) of gold nano rod solution and add to 1 mL solvent of your choice (play with the dilution as well). After dilution, drop cast it on the slide or silicon wafers and dry it in a desiccator. Let me know of the SEM results.

Hi Mudassir Khan, It is owing to the presence of an excess amount of surfactant that actually causes such issue. Dilution won't help unless the excess surfactant is discarded. I would suggest that centrifuge (RPM 5000- 7000; 10-15 min) the Au NRs solution twice (or trice) and drop-cast the concentrated Au NRs sediment on Silicon wafer (glass slide does not work as it is not electrically conducting) and subsequently, dry it either under vacuum or IR lamp. And please capture the SEM images at the periphery region of the drop-cast where good self-assembly of Au NRs can be found. Please let me know if you need any further help.
