I have a question about intracellular dye delivery by intophporesis. I have a setup consisting of microelectrode connected to an electric circuit where I measure the membrane potential. I would like to include a dye into microelectrode to label impaled cells (like PI and LY). I pass current through the microelectrode. My question is - what should be the relation between polarity of the dye and the current? For example for a positively charged PI should the current be positive or negative to facilitate it's diffusion/transfer in the cell?
