I have a trouble with high glucose concentration in DMEM. I am not sure how to make a DMEM with 50 mM glucose concentration from low glucose DMEM (5.5 mM)
The only way to reach high glucose concentration is to externally add sterile glucose solution. Please make concentrated glucose solution viz. 1M solution; and use it in your low glucose solution to reach 50mM concentration. Please calculate, you will have to add around 5 ml/lt DMEM solution.
Or, if you want to avoid all this; order high glucose DMEM from Gibco or Sigma

Vy, to keep the volume change to a minimum, use a 2M filter sterilized stock of glucose solution. To raise the concentration from 5.5mM to 50mM, you will need to use simple but correct Math.
Alternatively, you can add solid glucose to DMEM, change in volume will be much less this way but you will need to filter sterilize the entire medium.
You realize that 50mM glucose is rather unusually high concentration and will significantly increase osmolarity of your medium.
