I was trying to follow your protocol to monitor the enzymatic activity a GDH enzyme but I had some problems.
My question is: Did you use a solution of glutamate or a powder? If it's a solution, could you please provide me with the company name? So far I have only been able to locate glutamate in powdered form.
If it's a powder, could you please tell me what steps you took to dissolve the glutamate? At pH 7.5, the powder seems insoluble at concentrations higher than 30mM. While I can get the powder to dissolve at a lower pH, the acidity has an effect on enzymatic activity. If I try to gradually raise the pH back to 7, the glutamate precipitates.

you must heat it up to 50 C as its solubility in water increases with temperature (Pertzoff, JBC,1932).

Sodium glutamate is highly soluble in water (739g/L at 25°C which is roughly 4M) according to its pubchem data. Make sure you are using a glutamate salt.
