Does anyone have experience with lysosome isolation and purification? Since the pH conditions are different inside and outside lysosome, are the isolated lysosomes stable under buffer solutions? Any suggestions about the pH of the buffers for isolation and storage?

Concerning the pH of isolating buffer it should be ideally neutral similarly to cytoplasmic pH that lysosomes feel in the cells. If the point is keeping acidic internal pH after isolation what you need to provide is an ATP-regerating system to make sure that v-ATPase continues working.
