I am using "pHrodo™ Red E. coli BioParticles™ Phagocytosis Kit for Flow Cytometry" to access phagocytosis in neutrophils on perepheral whole blood.

As per the protocol i followed all the steps:

tube 1: Blood on Ice

tube 2: Blood+ beads on ice

Tube 3: Blood on 37 degree

Tube 4: Blood + beads on 37 degree

The tubes on ice are negative control for phagocytosis and activity is expected to be null.

I found 0% positivity in tube 1, tube 2 and tube 3, while 89% in tube 4 in a control.

Medians of tube 1 = 788, tube 2= 800, tube 3 = 750 and tube 4 is 1800.

How to interprete this data and report it.

Thanks in advance.
