Hi, Hala Altaher Inducing inflammatory response using LPS could be complicated as there are so many different serotypes. I'm not sure what inflammation marker you screened, but the cells react distinctively among different LPS strains. So, you may want to check some papers to see if the testing panel matches the stimuli. Also, some studies used conditioned medium from immune cells to induce inflammation in cultured intestinal epithelial cells.

Did you check the expression of TLR4 and TLR2 in your cell lines? Passaging and cultivation conditions can lead to the loss of some membrane receptors.

hello Yuning Hou , I have tested NO production through Griess reagent assay, also qPCR for IL6, IL1B, TNF-alpha but all showed no difference compared to untreated. I ordered LPS based on what has been used by some papers but still I see no response. I will try conditioned medium this is a good suggestion. thank you!
