I'm studing UVB-induced activation of inflammasome, I've tried the method of LPS+UVBR : I seeded the J774A.1 cell in a 24-well plate, 16hr later, I pretreated cells with LSP for 3.5hr(culture media was changed to Ultrafectin), then put cells under a UVB lamp for different time, but 5hr later, there's even no significant pyroptosis happend(observed by microscope), and there's no mature IL-1beta secreted(assay by WB).
Could anyone tell me the exact way to induce inflammasome by UVB in macrophages? especially the UVB dosage(mJ/cm2), radiation time, irradiance(mW/cm2) ?
