I want to observe interaction between plant proteins under DNA damage conditions using BiFC, so I need to perform DNA double-stranded breaks prior to (or maybe after) transient assay. Is there any suggestion of the method to use chemicals/constructs/irradiation.... to efficiently induce DSB in plants, and which plant materials (such as mature leaves, tissue culture, or cell culture, etc.) would bring out the best results? Any idea is deeply appreciated.
